  • May be induced if necessary.
  • 5-10 mL (minimum volume 2 mL) body fluids, tissues, exudates, cutaneous, gastric, etc.
  • 50 mL urine
  • 10 mL CSF (minimum volume 6 mL)
  • Blood cultures for AFB: Two 7 mL green top tubes.
Stools for AFB must be approved by the microbiology director.

Swabs are unacceptable for culture.

Storage/Transport Temperature

Internal: Deliver to lab immediately.
Offsite: Date and refrigerate specimens. Specimens must be delivered to the laboratory as soon as possible (no more than 2 days) for optimal recovery of AFB.

Stability (from collection to initiation)

Refrigerated: 48-72 hours.

Remarks

Test includes Mycobacterial (AFB) culture and stain. Physicians will be notified of all positive cultures and/or stains. Negative cultures will be reported after 8 weeks.

Sensitivity testing will be performed on M. tuberculosis by an outside laboratory. Sensitivity testing on MOTB must be approved by Infectious Disease.

Respiratory specimens: Since 5-10 mL sputum (not saliva) is required, specimens may be collected over 1-2 hours. Refrigerate if immediate delivery to lab is not possible.

Urine specimens: No AFB stain is performed due to the presence of saprophytic mycobacteria in genital secretions of normal patients.

CSF specimens: No AFB stain is performed due to extremely low sensitivity of the test.

Collect

In a sterile container:

  • 5-10 mL (minimum volume 2 mL) respiratory specimens. Submit three early moring specimens on three consecutive days.  Early morning specimens are preferred.
  • To expedite the patient's discharge, submit 3 sputa, one in the early morning and the remaining two at least 4 hours apart in one 24 hour period.  All three specimens will be processed the following day.
  • May be induced if necessary.
  • 5-10 mL (minimum volume 2 mL) body fluids, tissues, exudates, cutaneous, gastric, etc.
  • 50 mL urine
  • 10 mL CSF (minimum volume 6 mL)
  • Blood cultures for AFB: Two 7 mL green top tubes.
Stools for AFB must be approved by the microbiology director.

Swabs are unacceptable for culture.
